I've successfully installed WordPress in advanced mode with a new database.
What I'd like to do is install a second instance in a new directory. Should I also create a new database or can I use the existing one? Thanks for any input.

You could use the same database, but why? I really like to keep my installations separate. With a separate database, it's easier to dump and restore just the one installation.

Create a new database too. If you actually wanted to have one installation of WordPress but have it run separate blogs then see http://mu.wordpress.org/
